A 95 Peptidstørrelsesfordeling – Validering ny kolonne

Abstract

For analysen ‘A 95 peptidstørrelsesfordeling’ benytter BioLab Size Exclusion Chromatography (SEC) kolonnen Superdex Peptide. Ved ønske om å kjøpe inn en ny kolonne av den samme typen ble det oppdaget at denne var tatt ut av produksjon. Fra leverandøren sin side var det anbefalt en kolonne som avtager, Superdex Increase. Etter en validering der de to kolonnene ble sammenlignet ble det konkludert med at kolonnen Superdex Increase kunne brukes som en avtager for Superdex Peptide. Dette medførte derimot en noe større måleusikkerhet, og en innskrenkning i måleområdet fra ‘<200 – 20 000<’ til ‘<200 – 15 000<’.

In the analysis ‘A 95 Molweight Distribution of Peptides’ BioLab use the Size Exclusion Chromatography (SEC) column Superdex Peptide. After a request for a new column of the same type it was informed that the column was no longer produced. The supplier recommended a replacement column, Superdex Increase. After a validation where the two columns was compared, it was concluded that BioLab could use Superdex Increase as a replacement for Superdex Peptide. This lead to a slight increase in the uncertainty for the methode, and a restriction in the measuring area. From ‘<200 – 20 000<’ to ‘<200 – 15 000<’.
